Abstract:
In order to provide technical reference for artificial breeding of Dendrobium officinale situated areas north,taking purchased tissue culture seedlings and potted seedlings as test materials,different concentrations of 6-BA and NAA were used to induce axillary buds,protocorms and adventitious roots to establish regeneration system,rapid propagation tissue culture seedlings of Dendrobium officinale.The results showed that the explants should be disfected by 70% alcohol for 20 seconds and 0.1% mercuric chloride for 8 minutes.MS+6-BA 3.0-5.0 mg?L-1+NAA 0.1-1.0 mg?L-1 was suitable for axillary buds differentiation and proliferation.The differentiation rate was 92%-100%;1/2MS medium with 6-BA 2.0 mg?L-1+NAA 0.5 mg?L-1+ mashed potato 15% was suitable for protocorms proliferation with a proliferation multiple of 7.7.In 1/2MS medium with NAA 0.3 mg?L-1,the rooting rate was 100%.Transplanting with pine needle soil,the survival rate was 83%.
