Abstract:
The annual stem segments of the new colored tree cultivar Melia azedarach‘Meiren’ were used as experimental material to study the effect of different concentrations of 6-BA,IBA and NAA on its tissue culture so that to establish an efficient rapid propagation system of tissue culture for Melia azedarach‘Meiren’.The results showed that the explants were disinfect with 0.2% mercury for 15 minutes after sterilizing for 10 minutes by 1 g?L-1 carbendazim and 5 minutes by 300 mg?L-1carboxy benzyl.The contamination rate was only 32%.The suitable culture medium for adventitious bud proliferation was MS+6-BA 0.3 mg?L-1+IBA 0.20 mg?L-1 with the proliferation coefficient was 4.76.While 1/2MS+IBA 0.10 mg?L-1+NAA 0.05 mg?L-1 was the ideal culture medium for rooting.The rooting rate could reach 90.8%.The survival rate was 90% when the plantlets were transplanted in the substrate of grass carbon∶perlite=2∶1.This study established a set of perfect rapid propagation systemof tissue culture for the new colored tree cultivar of Melia azedarach,which had the important theoretical and practical significance to the protection of germplasm resources,propagation,spreading and genetic improvement of the new cultivar.
