Abstract:
Using Hypoxylon HYP-GT01 as test material,by adjusting the composition of culture medium,pH and light intensity,the morphological characteristics,growth rate and melanin yield of hyphal under different culture conditions were analyzed in order to find out the main factors affecting the hyphal growth of Hypoxylon.The results showed that the growth rate of the hyphae was the fastest in the culture medium.The dense edge of the hyphae was neat.The melanin was produced,and the yield was 0.42 g?L-1,was the highest.The hypha growth was slow in the low-nitrogen medium,but the hyphae were thick,and the melanin could be produced up to 0.34 g?L-1.The carbon source wasn′t enough for the filament to be thin and sparse,and it didn′t produce melanin.High inorganic salt content and the addition of Tween 20 could inhibit mycelium growth and produce a small amount of melanin.The pH level in a certain range had little effect on the growth and melanin.Different light conditions were not only affect the growth rate of mycelium,but also the production of melanin.
