Abstract:
The genetic diversity of 77 Forsythia suspensa germplasm resources collected from 12 areas of Henan and Shanxi Provinces was analyzed by EST-SSR molecular marker technique,in order to explore the genetic similarity of Forsythia suspensa in different habitats and its key influencing factors.The results showed that 14 pairs of polymorphic EST-SSR primers with clear amplified bands and good stability were screened from 60 pairs of primers.14 pairs of primers amplified 370 bands in 77 samples,including 368 polymorphic bands,and the percentage of polymorphic sites was 99.46%.The genetic distances of the 12 areas ranged from 0.020 3 to 0.092 2,and the average genetic distance was 0.055 8.According to genetic similarity coefficient,12 populations were clustered into 5 groups at 0.935 sites and 77 samples were clustered into 5 classes at 0.935 sites.The result of individual clustering was similar to that of origin.The analysis of clustering results showed that the genetic similarity of Forsythia suspensa had no significant correlation with geographical distance,which was mainly related to growth environment and altitude,etc.The experimental results reveal the key factors of genetic similarity of Forsythia suspensa in different habitats,which lays a foundation for the identification of Forsythia suspensa susceptibility and the protection and development of Forsythia resources.
