Abstract:
The mutagenic effect of EMS on the seeds of radish was studied by soaking seeds with different concentration of EMS(0,0.2%,0.4%,0.6%,0.8%,1.0%,2.0%(V/V)).In order to establish a good foundation for the breeding and utilization of male sterile line of radish by obtaining the sterile material of Huangzhou radish.The results showed that in the aspect of germination rate and root length,the growth of radish was inhibited with the increase of EMS concentration (from 0.2% to 2.0%).So the inhibition of EMS to Huangzhou radish was obvious.According to the selection criteria of 50% lethal concentrations,the suitable EMS concentration of Huangzhou radish was 0.4%-0.8%.The seed of Huangzhou radish was treated with 0.5% EMS.It was found that the main agronomic characters of the treatment group were thickened by compared with control group.Furthermore,the male sterility in the Huangzhou radish mutation group was found and it was very stable.The results showed that the anther was obviously wrinkled,curled and brown,and eventually the pollen abortion was caused by disintegration of the inter wall of pollen by morphological and cytological observation.It also showed that it could be enlarged to the corresponding size bands in the male sterile line of Huangzhou radish,while the band was not amplified in normal line by detection of molecular marker ORF138-F/R in radish.
