Abstract:
Abstract:Theeffectsofdifferentbasicmedium,exogenousadditives,addingpolyacrylamide,differentcultureconditionsandshootsofdehydrationonvitrificationshootsrecoverybydifferentvitrifiedseedlingsofLyciumbarbarumasexplantsfortissueculturewerestudied.Themainpurposeofthestudywasexplorethemeasuresofwhichmakethevitrificationshootsrecovery.Theresultsshowedthat1/2MS+6-BA05mg?L-1+activatedcarbon05g?L-1wasthebestmediumforthemildvitreousseedlingsrecoveryofLyciumbarbarum.ItwassuitableforthemoderatevitrifiedseedlingsrecoveryofLyciumbarbarum1/4MS+6-BA05mg?L-1+activatedcarbon10g?L-1.Whenpolyacrylamide8g?L-1and16g?L-1wereadded,therecoveryrateofmildvitreousandmoderatevitrifiedseedlingsofLyciumbarbarumwasthehighest.ItnotonlycouldsignificantlyinhibitthegenerationofvitrficationofLyciumbarbarum,butalsohadsignificantreversaleffectwithvitrifiedtissue-culturingplantlets,whichgrewwellafterreverse.Afterdehydrationtreatment,thevitrifiedseedlingswereobviouslyrecovered.Therecoveryrateoflightvitrifiedseedlingsofdehydrated2dayscouldreach7761%.Withtheincreaseofthenumberofdaysofdehydration,therecoveryrateofmediumvitrifiedseedlingsofLyciumbarbarumwasgraduallyincreased.Whenthenumberofdehydrationdaysreached3days,thehighestrecoveryratecouldreach7124%.Enhancingmembranepermeabilityandlightintensitycouldalsoraisetherecoveryrateofvitrifiedseedlings.
