Abstract:
A pre-column derivation coupled HPLC analysis was applied to identify and characterize polysaccharides of Ophiocordyceps sinensis and relative cultivated mycelium from different origins.The polysaccharides extracted from O.sinensis were hydrolyzed partially with trifluoroacetic acid and derivated by 1-phenyl-3-methyl-5-pyrazolone (PMP).The monosaccharide composition fingerprint was obtained by HPLC on a Agilent Eclipse XDB C18(250 mm×4.6 mm,5 μm)column with phosphate buffered solution (0.1 mol·L-1,pH 6.7) and acetonitrile (80∶20,v/v) as mobile phase at the flow rate of 1 mL·min-1.The column temperature was at 25 ℃.The results showed that the similarity of 12 batches of O.sinensis was more than 0.992.There were some differences between O.sinensis and cultivated mycelium,with the similarity value range from 0.708 to 0.974.The result of Cluster analysis (CA) largely agreed with the similarity evaluation,which distinguished O.sinensis from cultivated mycelium.The polysaccharides fingerprint by HPLC combined with CA were simple and reliable,which could precisely differentiate O.sinensis from cultivated mycelium and were useful for quality evaluation of O.sinensis.
