Abstract:
Based on the genome sequence,mitochondrial intermediate peptidase encoding gene (mip) was cloned from two strains YSG and AC0007 using long-range PCR method,and the genetic characterization in Agrocybe aegerita strains population were also investigated to test the applicability of them as the molecular mark for polarity identification.The results showed that mip was co-segregated with A mating locus and therefore it could be used for the molecular for the cloning of the latter.Sequence analysis showed C terminal of MIP protein encoding region was highly conserved,and the special primer pairs were designed for this region could amplify the mip fragments in all tested strains.Additionally,the variable encoding regions in C terminal could be used as the targeting site for the A mating locus identification,and it provided an important basis for genetics and breeding of edible mushroom.
