- Abstract:
- Based on the genome sequence,mitochondrial intermediate peptidase encoding gene (mip) was cloned from two strains YSG and AC0007 using long-range PCR method,and the genetic characterization in Agrocybe aegerita strains population were also investigated to test the applicability of them as the molecular mark for polarity identification.The results showed that mip was co-segregated with A mating locus and therefore it could be used for the molecular for the cloning of the latter.Sequence analysis showed C terminal of MIP protein encoding region was highly conserved,and the special primer pairs were designed for this region could amplify the mip fragments in all tested strains.Additionally,the variable encoding regions in C terminal could be used as the targeting site for the A mating locus identification,and it provided an important basis for genetics and breeding of edible mushroom.
