Abstract:
Young stems and young leaves of Matricaria chamomilia were used as explants,by adopting the method of single factor experiment,the influence of different disinfection measures and different hormone concentrations on callus induction of M.chamomilla was studied,in order to establish M.chamomilla regeneration system and genetic engineering methods.The results showed that young stems were the best explants for tissue culture with a high level of callus forming rate of 90%.The best condition of sterilization with young stems was as follow:fresh young stems wash under sterile water for 30 minutes,followed with 75% alcohol for 10 seconds and then 3 5% sodium hypochlorite for 10 minutes,the induction rate was as high as 86 7%,pollution rate was as low as 13 3%;the best medium for callus induction was MS + 6-BA 1 5 mg?L-1 + NAA 1 2 mg?L-1.
