Abstract:
The factors affecting callus initiation from magrove plant Bruguiera gymnorrhiza was investigated with the young leaves as explant.Sterilization conditions,medium components,kinds of additive,different concentration of plant growth regulator combination were tested and callus induction rate was calculated.The results showed that 75% alcohol 30 seconds with 2% (volumn fraction w/v) NaClO 20 minutes was the best condition of surface sterilization;amino acid (AA) medium was basic medium;browning rate was inhibited by heat-shock at 45 ℃ and ascorbic acid;AA medium+0.50 mg?L-1 2,4-D+0.50 mg?L-1 TDZ+1.00 mmol?L-1 vitamin C or AA medium+0.50 mg?L-1 2,4-D+0.50 mg?L-1 CPPU combination were optimal callus induction medium,in which over 50% of callus induction rate was obtained.Therefore,explant browning prevention and application of CPPU or TDZ as cytokinine were the key to success in callus induction from Bruguiera gymnorrhiza.
