Abstract:
It was experimented to screen suitable temperature for Cordyceps militaris mycelium.After that,liquid medium was optimized by response surface method.Finally,using the optimized conditions,the mycelium was staticly fermented 10 days after 6 days shaking-flask culture,compared with 6 days shaking-flask culture,in order to understand production of Cordyceps militaries mycelium under different culture conditions and the effect of static fermentation on cordycepin production.The results showed that optimal conditions by one-factor experiment were dark at 25℃.Optimal liquid medium by response surface was sucrose 4.34%,yeast extract 3.06%,FeSO4 0.027%,KH2PO4 0.2%,(NH4)2SO4 0.04%,MgSO4 0.13%,vitamin B1 0.08%,ZnSO4 0.06%.The yield of mycelium by cordyceps militaris in the optimal medium increased 1.73 times compared with that in initial medium and reached 2.568 g/100mL,while the yield of mycelium reached 3.389 g/100mL with two-stage culture.And the results of two-stage culture showed that the yield of cordycepin increased 1.56 times more that in initial medium and reached 862.893 μg/mL,compared with 568.329 μg/mL of only 6 days shaking-flask culture.
