Abstract:
The detection methods of four endogenous hormones [gibberellin acid(GA3),indole-3-acetic acid(IAA),abscisic acid(ABA),zeatin(ZT)]in roots of bananas by HPLC with UV detector were established.The separation was performed on an Agilent ZORBAX SB-C18 column (250 mm×4.6 mm,5 μm).The optimum chromatographic conditions were as follows:methanol∶acetonitrile∶phosphate buffer solution(pH 3.5)=15∶20∶65 as the mobile phase at 1 mL/min with an injection of 20 μL for GA3,IAA,ABA at 210 nm;methanol∶acetonitrile∶phosphate buffer solution(pH 3.5)=15∶15∶70 as the mobile phase at 1 mL/min with an injection of 20 μL of ZT at 265 nm.A column temperature of 35℃ was optimum for the separation.The results showed that the recoveries were all high,and this method was rapid and perfect.
