Abstract:
Taking diseased plants with typical symptoms as test materials,pathogen isolation was carried out by means of diseased tissue isolation method.Koch′s rule was used to prove the pathogenicity of fungal strain,in order to explore the main pathogens of Ligusticum jeholense root rot disease and its biological characteristics.The results showed that based on the morphological observation and sequence analyses of translation elongation factor EF-1α, Fusarium solani was identified as the pathogen.The biological characteristics showed that different media had significant effects on the growth and mycelial density of pathogen.The suitable medium for Fusarium solani growth was PDA,the colony diameter reached 85 mm in 7 days,and the colonies were dense.The growth of mycelia with sucrose as carbon source was faster than that with soluble starch,and the ability of pathogen to utilize lactose was poor.The most suitable nitrogen source was beef extract,followed by urea and potassium nitrate.The suitable temperature for the mycelium growth was 20-30 ℃,the optimum temperature was 28-30 ℃.The optimum pH was 6.The light treatment had an influence on the growth of mycelium,suitable light condition for the mycelium growth was 24 hour continuous dark.
