Abstract:
Taking hybrid orchid ‘Hongmeiren’×Cymbidium kanran ‘Yangguifei’ of rhizomes as experimental material,using L9(34) orthogonal experiment method,the effects of 6-BA,NAA,TDZ on different photoperiod and basic culture medium of rhizomes proliferation,leaf bud differentiation and flower bud differentiation were studied,in order to explore the hybrid laingen stems proliferation and differentiation of suitable conditions.The results showed that the best formula of rhizome proliferation,2.0 g?L-1 Huabao 1+1.0 g?L-1 Huabao 2+1.0 mg?L-1 TDZ+1.0 mg?L-1 NAA+0.1g?L-1 activated carbon+40.0 g?L-1 sucrose+7.0 g?L-1 agar+6.0 g?L-1 proteinpeptone+40.0 g?L-1 banana+40.0 g?L-1 potato;the proliferation coefficient was highest at 12 hours/12 hours (light/dark) and the fastest growth rate at 10 hours/14 hours (light/dark).The best formula of leaf bud differentiation,2.0 g?L-1 Huabao 1+1.0 g?L-1 Huabao 2+5.0 mg?L-1 6-BA+1.0 mg?L-1 NAA+0.1 g?L-1activated carbon+40.0 g?L-1 sucrose+7.0 g?L-1 agar+6.0 g?L-1 peptone+50.0 mg?L-1 coconut milk.In MS+5.0 mg?L-1 6-BA+1.0 mg?L-1 NAA+0.1 g?L-1 activated carbon+40.0 g?L-1 sucrose+7.0 g?L-1 agar+6.0 g?L-1 proteinpeptone+40.0 g?L-1 banana+40.0 g?L-1 potato,the rhizome produced flower buds.
