Abstract:
The disease plants of gerbera root rot were used as the experimental materials,pathogen species were isolated and purified from the gerbera root rot by the tissue isolation method,then the pathogenicity of the isolates to gerbera plants were studied,finally the pathogen species were identified by using morphological and molecular systematics method based on rDNA-ITS and TEF-1α sequences,in order to make clear the fungal pathogen species of thegerbera root rot disease.The results showed that a total of 8 pathogenic strains of FZJf 1-1,FZJf 1-2,FZJf 1-3,FZJf 2-1,FZJf 2-2,FZJf 2-3,FZJf 3-1 and FZJf 3-2 were isolated from severely rot roots;pathogenicity test results showed that all the 8 strains were pathogenic to gerbera seedlings and could cause typical leaf wilt and root rot symptoms,among which strains FZJF 1-1 and FZJF 1-3 had the strongest pathogenicity,the plants developed disease after 3 weeks inoculation,followed by strain FZJF 2-1,the plants developed disease after 5 weeks inoculation,while the other strains were less pathogenicity;6 strains of FZJf 1-1,FZJf 1-2,FZJf 1-3,FZJf 2-1,FZJf 2-3 and FZJf 3-2 were identified as Fusarium oxysporum,one strain of FZJf 2-2 was identified as F.solani and one strain of FZJf 3-1identified as F.proliferatumby morphological and molecular phylogenetic characteristics.F.oxysporum species were the dominant pathogens,and F.solani and F.proliferatum were weak pathogens.
