Abstract:
A wild Lentinula edodes from Papua New Guinea was used as test material,we used sequence analysis of ITS,LSU and SSU for molecular identification,and used single factor tests and orthogonal tests to explore its mycelial biological characteristics.In order to clarify the taxonomic status of the wild Lentinula edodes and provide a reference for the artificial cultivation and further research of Lentinula lateritia.The results showed that the length of the ITS sequence was 725 bp,the length of LSU sequence was 934 bp,and the length of SSU sequence was 1 100 bp,which had the highest similarity with Lentinula lateritia.The strain was identified as Lentinula lateritia.The results of biological characteristics showed that the optimal carbon source for the growth of wild Lentinula edodes mycelial was fruticose,the optimal nitrogen source was beef extract,the optimal pH was 4.0,and the optimal growth temperature was 25 ℃.
