Abstract:
Based on the genome and transcriptome data of Flammulina filiformis,this experiment identified an expansin analog encoding gene using online software BLAST.It was named [STBX]fvexpl2[STBZ] and identified its gene structure.The phylogenetic relationship between [STBX]fvexpl2[STBZ] and basidiomycetes,ascomycetes and plants was analyzed by constructing evolutionary tree.The results showed that four exons and three introns contained in the dilatant protein analogue,and the length of coding sequence was 438 bp,molecular weight 15 746.88 Da,isoelectric point 4.93.The construction of evolutionary tree showed that [STBX]fvexpl2[STBZ] and basidiomycetes were in the same subclan and had close relationship.According to the real-time fluorescence quantitative PCR results,we could understand that [STBX]fvexpl2[STBZ] was much higher than that in the cap,primordium and mycelium,and it had a higher expression level in the fast elongation region of the stipe compared with the slow elongation region.Therefore,[STBX]fvexpl2[STBZ] might play an important role in the elongation of the stipe.
