Abstract:
The effects of different basic medium,hormone combinations,sucrose concentration and light duration on callus induction,adventitious bud induction,adventitious root induction and root tuber induction were studied by using Dahlia pinnate ‘Filogo’ stem segments as explants,in order shorten the production cycle of root tuber.The results showed that the callus induction rate,browning rate,contamination rate and survival rate were 54.9%,44.8%,26.6% and 69.6% respectively on MS+6-BA 2.0 mg·L-1+NAA 0.1 mg·L-1+sucrose 30 g·L-1+agar 6 g·L-1.The optimum medium for subculture was MS+6-BA 2.0 mg·L-1+NAA 0.01 mg·L-1+sucrose 30 g·L-1+agar 6 g·L-1,the proliferation coefficient was 7.8,and the callus induction rate was 22.9%.The optimum medium for rooting was WPM+NAA 1.0 mg·L-1+6-BA 0.1 mg·L-1+sucrose 60 g·L-1+agar 6 g·L-1+carbon powder 2 g·L-1.The optimum illumination time was 6 hours per day,the rooting rate was 98.5%,and the roots were dense and robust.The optimum material for root induction was bottle seedling with roots.The rate of root tuber formation with initial morphology was 56.8%.
