Abstract:
Using tomato ‘CM966’ disease strain as test material,the suspected tomato disease strain collected by Baotou city was identified by PCR method and morphological identification method,and the phylogenetic tree was constructed to determine its taxonomic status;17 tomato lines and cultivars were tested with specific PCR amplification and using Rcm5.1 with the highest contribution rate in the QTL locus associated with tomato canker disease,and field inoculation test was conducted to explore tomato canker resistant species resources.The results showed that the suspected canker disease strains from four different tomato plantations all carried tomato canker.After BLAST comparison,the isolates of 4 pathogens were more than 90% identical to the reported Clavibacter michiganensis subsp.michiganensis (Cmm) in GenBank,including 1 pathogen.The isolate sequence was significantly different from other strains;17 tomato lines and cultivars were tested,and none of the resistance genes at the Rcm5.1 point in the QTL locus was carried.However,field vaccination trials showed that three strains ‘Shandong Holland’,‘Sheng 490’ and ‘Amethyst’ showed high resistance,with disease indices of 2%,4%,and 4%,respectively.‘160’‘CM966’‘T54’ and ‘Provence’ showed high susceptible,and the disease index was greater than 50%.
