Abstract:
To explore the influences of temperature (5-35 ℃) and pH (3-9) on mycelial growth,sporulation ability and toxins production of Valsa mali,the strain LXS080601 with high virulence was studied by using growth rate method and high performance liquid chromatography (HPLC).The results showed that the strain LXS080601 was able to grow at a temperature range from 5 ℃ to 35 ℃ and at a pH range from 3 to 9.The optimal temperature and pH for LXS080601 ranged from 25 ℃ to 30 ℃ and from 4 to 6,optimal temperature and pH for LXS080601 ranged from 25 ℃ to 30 ℃ and from 4 to 6,respectively.pH also had significant effects on sporulation of V.mali.The optimal pH for LXS080601 sporulation ranged from 5 to 6,and the number of conidia per plate reached to 315.In apple bark extract medium,the optimal temperature for toxins production was from 20 ℃ to 25 ℃,and the total toxins amount was up to 128.06 μg?mL-1.Five kinds of toxins were produced by strain LXS080601 from 10 ℃ to 35 ℃,however,only three kinds of toxins were detected at 5 ℃ and the total toxins amount was only 8.46 μg?mL-1.In addition,the optimal pH for toxins production was from 3 to 6,while at pH 9 only four kinds of toxins were detected and the total toxins amount was the lowest.
