Abstract:
The Morchella collected from 8 different regions was used as the test material.The separation column was uncoated quartz capillary (50 μm×48 cm);the running buffer 10 mmol?L-1 borax buffer (pH 9.21);10 mmol?L-1 boric acid buffer detection wavelength 214 nm;separation voltage 20 kV;temperature 25 ℃;pressure 0.5 psi injection 10 seconds,running time 30 minutes electrophoresis conditions,in order to establish a quality control method based on capillary electrophoresis technology of Morchella fingerprint.On the basis of method validation,the fingerprints of more than 12 common peaks were established.The No.10 peak was used as the reference peak of the fingerprint of the strain,and the quality evaluation and identification of 8 species of Morchella were carried out by similarity evaluation,cluster analysis and principal component analysis.The results showed that the results of this study were consistent with other molecular identification methods.The method had simple sample processing,easy to realize chromatographic conditions,no toxic and harmful solvent,no harm to the environment,good stability and reproducibility,and was suitable for the identification and control of the quality of the morel.
