Abstract:
To develop new tissue culture and rapid propagation systems of Tripterygium wilfordii Hook.f.,the young shoots were used as explants to induce axillary buds germination.Germinated axillary buds were cut for root inducing and elongation of stems and leaves.The elongated stems were excised into budded stem segments,from which roots were induced in a specific culture medium to achieve rapid plant proliferation.The results showed that the best sterilization effects were achieved when the explants were soaked in 75% alcohol for 30 seconds,washed three times with sterile water,soaked in 0.1% mercury liter solution for 10 minutes.The most effective medium for axillary bud germination was MS medium with 1.5 mg?L-1 6-BA+0.2 mg?L-1 NAA.The efficacy could reach 98.3%.The optimum medium for inducing rapid rooting of stem segments was 1/2MS medium with 0.2 mg?L-1 NAA+0.1 mg?L-1 IAA+0.03% activated carbon.The optimal root-inducing condition was air suspension culture with the lower stem ends of ecology at about 1 cm height from the medium at (23±1)℃,and the humidity being 70%±5% by the 12 h/12 h light/dark culture.The germination efficacy of adventitious roots reached 91.5%.In this study,a tissue culture system for rapid propagation of Tripterygium wilfordii seedlings was established.With this simple method,the monthly proliferation coefficient could reach 3.8.Cutting and transferring budded stem segments to root-inducing medium could realize rapid proliferation of stems and leaves and rooting at the same time.
