Abstract:
JUNCAO-cultivated Ganoderma lucidum spent mushroom substrate was used as test material.The content of crude polysaccharides,triterpenes and amino acids and other ingredients in the water extract of Ganoderma lucidum spent mushroom substrate (SMSG-WE) was determined by using national standards,agricultural industry standards,and the extraction process of crude polysaccharides from SMSG was optimized by orthogonal test in the laboratory,the effects of different solid-liquid ratio,extraction temperature,extraction time and times on the yield of polysaccharide from Ganoderma lucidum spent mushroom substrate were studied to obtain the optimal extraction conditions.On this basis,the pilot production process of SMSG-WE was optimized.The results showed that the contents of crude polysaccharides,triterpenes and amino acids were 24.16%,0.82% and 6.50%.In laboratory the optimum extraction condition of polysaccharides was solid-liquid ratio 1∶20 g?mL-1,extracting temperature 100 ℃,extraction time 4 hours,extraction times for 3 times,under the best conditions,the polysaccharide yield is 4.45%;pilot extraction conditions of SMSG-WE were determined as follows,extraction for 3 times,for the first time the solid-liquid ratio 1∶10 g?mL-1,heated to 100 ℃ for 2 hours,second time solid-liquid ratio 1∶8 g?mL-1,heated to 100 ℃ for 2 hours,third time solid-liquid ratio 1∶6 g?mL-1,keep 1.5 hours after heated to 100 ℃,the average obtained at a rate of 20.03%.
