Abstract:
This study was designed to screen for the optimal medium for root growth of (Pterocephalus hookeri (C.B.Clarke) Hock.).The sterile seedling of Pterocephalus hookeri was used as the research object.The roots were obtained by infecting the leaves with Agrobacterium tumefaciens C58C1 (pRiA4) to study the effects of different infestation conditions on the number of hairy roots.The effects of five mediums (MS,1/4MS,B5,1/2B5 and WPM) on the root biomass and content of oleanolic acid and vrsolic acid for Pterocphalus hookeri were veriflied by the determination of fresh weight、dry weight、oleanolic acid、ursolic acid content in liquid culture.The results showed that the induction rate of Pterocephalus hookeri was the highest (12%) for infect 8 minutes,co-culture 1 day,and the root biomass was the highest in 1/4MS liquid medium.The fresh weight was 9.210 g and the dry weight was 0.529 g.The fresh weight of B5 medium was 5.270 g and the dry weight was 0.385 g.The roots of oleanolic acid and ursolic acid content were the highest in B5 medium,which were 2.314 mg?g-1 and 3.213 mg?g-1,respectively.1/4MS was the lowest,indicating that B5 was the best medium for root growth and production of Pterocephalus hookeri.
