- Abstract:
- In this study,four Kentucky bluegrass varieties were used as materials for the screening of appropriate sterilization and callus induction methods with the mature seeds as explants,in order to supply reference for bluegrass use.The results showed that,the optimal sterilization method was to treat the seeds by 1% NaClO for 7 minutes followed by 4-5 times of washing by distilled water.Subsequently,the seeds were immersed with sterile water for 12 hours.Right before inoculation,the seeds were again washed with 75% ethanol for 40 seconds and then with sterile water for 4-5 times,followed by 10 minutes sterilization by 1% NaClO.The four Kentucky bluegrass varieties possessed different genotypes as well as significantly different frequencies of callus induction (P<0.05).The optimal culture medium was MS+1 mg·L-1 2,4-D+0.1 mg·L-1 6-BA for Kentucky bluegrass ‘Midnight II’,MS+6 mg·L-1 2,4-D for Kentucky bluegrass ‘Park’ and ‘Black Tiger’,and MS+1 mg·L-1 2,4-D for Kentucky bluegrass ‘Snow Wolf’,respectively.
