- Water extract of wild apricot leaves was used as material,and lipopolysaccharide (LPS)-induced mouse mononuclear macrophages RAW264.7 as the inflammatory cell model.Several methods were operated to detect the cell viability alteration and the expression change of inflammation related genes,including CCK8,Griess method,enzyme linked immunosorbent assay (ELISA) and RT-PCR,in order to study the anti-inflammatory effect of water extract of wild apricot leaves on RAW264.7 cell inflammatory model induced by LPS.The results showed that when the water extract of wild apricot leaves was co-cultured with 1 μg·mL-1 LPS in the concentration range of 0.5-2.0 mg·mL-1,it had no obvious effect on the proliferation of RAW264.7 cells;the treatment of LPS could significantly increase the secretion of NO and IL-6,but the treatment of water extract of wild apricot leaves could effectively inhibit the release of NO and the production of IL-6;and at gene level,the water extract significantly inhibited the mRNA expression of inflammatory factor IL-12p40,COX-2,IL-6 and TNF-α,in a concentration-dependent manner.
