- Abstract:
- A local line pepper root tip was used as plant material.Factors that affected chromosome preparation including sampling time,pretreatment reagents and time were analyzed,in order to optimize the chromosome preparation procedure and investigate the karyotype of line pepper and to provide a theoretical basis for studying genetic and phylogenetic relationships of different types of malabar spinach.The results showed that the percentage of the mitotic cells and metaphase cells reached maximum when sampled at 10:00;the best contraction,shape and dispersion of chromosomes were found at 3 hours 8-hydroxyquinoline pretreatment,which were easy for counting and karyotype.The karyotype formual was 2n=2x=24=22m+2sm,karyotype asymmetry belongs to 2A and the karyotype asymmetry index was 54.0%.This study established an optimal chromosome pretreatment for a local line pepper in Guizhou,and revealed its karyotypic characteristics from the cytogenetic aspects.
