Abstract:
The anthers of eight genotypes watermelon were used as test materials to investigate the effect of genotype and plant growth regulator on callus induction,and effect of dark culture and ascorbic acid on anther browning,in order to provide reference for the haploid breeding of watermelon.The results showed that 7 of 8 watermelon materials could form callus and the callus induction rate was from 3.9% to 35.6%.The response of different genotypes to hormones was different,and the callus induction of ‘P15’ was the highest in the D5 medium (MS+KT 1.0 mg?L-1+NAA 0.5 mg?L-1),and the rate of ‘M30’ and ‘M34’ was the highest in the D6 medium (MS+KT 1.0 mg?L-1+NAA 1.0 mg?L-1).The dark culture for 48 hours was the best to reduce the browning effect of watermelon anther.The addition of 1.0 g?L-1 ascorbic acid in the induction medium could reduce the rate of anther browning.
