Abstract:
Hericium erinareus 0605 was used as test material,the influence of enzyme system,enzymolysis time,enzymolysis temperature,mycelial age and shaking speed on the preparation and regeneration of protoplasts of Hericium erinareus 0605 were studied by single factors and orthogonal design experiments.The results showed that the optimal preparing and regeneration conditions of Hericium erinareus protoplasts were mycelium age 10 days,using 2.0% lywallzyme and 0.5% driselase,digesting at 32 ℃,shaking speed 120 r?min-1 for 2.5 hours.Under this conditions,the study established the foundation of protoplast fusion breeding of Hericium erinareus.
