Abstract:
The stem segments and tender leaves which were current-year branches of Prunus persica (L.) Batsch were used as explants.Using the single factor design,the effects of different disinfection methods on the inoculated of two explants contamination rate and callus formation rate,and the rate of callus formation under different hormone types and concentration ratio were studied,in order to lay a foundation for the regeneration system and genetic transformation of P.persica.The results showed that the optimal surface disinfection of the stem segments of P.persica was 75% alcohol 30 seconds+2.5% NaClO 10 minutes;the optimum medium for stem segments was MS + 6-BA 1.0 mg?L-1+IBA 0.5 mg?L-1+GA3 0.5 mg ?L-1.And the optimal surface disinfection of the tender leaves of P.persica was 75% alcohol 30 seconds+2.5% NaClO 6 minutes+0.5% NaClO 10 minutes;the optimum medium for tender leaves was 1/2MS+TDZ 1.5 mg?L-1+NAA 0.1 mg?L-1.In this study,the regeneration system from stem segments and leaves of P.persica was established,which laid the foundation for the genetic transformation of peach.
