Abstract:
Rubus biflorus Buch was used as test material,total genomic DNA was extracted by using improved CTAB method.The factors including primers,dNTPs,Mg2+ and annealing temperature were studied to establish optimized SCoT-PCR reaction system to ensure the stability of SCoT for Rubus biflorus Buch.The results showed that the optimized reaction system of SCoT-PCR was consisted as follows,in the total reaction system with amount of 20 μL,there were 0.125 mmol?L-1 dNTPs,0.15 μL Taq DNA polymerase,1.0 μmol?L-1 primers,0.625 mmol?L-1 Mg2+,40 ng template DNA,respectively.The optimal annealing temperature was 51 ℃.The findings suggested that the products′ bands were clear with high stability.
