Abstract:
The stem apex,stem,leaves and petiole of Spiraea×bumalda ‘Golden Mound’ were taken as explants for tissue culture in vitro.Different species and concentrations of hormones were added to MS medium during the different periods of first,subculture and rooting culture for establishing of regeneration system suitable for tissue culture of Spiraea×bumalda ‘Golden Mound’.The results showed that,medium for callus induction was MS+1.0 mg/L 6-BA+0.1 mg/L NAA+2.0 mg/L 2,4-D,medium for adventitious bud induction was MS+0.8 mg/L TDZ+0.10 mg/L NAA,medium for rooting was 1/2MS+0.25 mg/L 6-BA+1.0 mg/L NAA,medium for axillary bud proliferation was MS+2.0 mg/L 6-BA+0.2 mg/L NAA.
