Abstract:
Taking thirty-seven peony cultivars from Shandong as experimental materials,the DNA fingerprinting database was successfully constructed,and the genetic diversity was analyzed based on ten evenly distributed EST-SSR primer pairs with high polymorphisms and good repeatability.The results showed that,among the 37 varieties,10 primer pairs had 58 polymorphic locis,and 5.8 polymorphic locis were detected by each EST-SSR primer pair on an average with the range from 2 to 13.EST-SSR markers were suitable for using in construction of peony DNA fingerprint with polymorphism differences between the selected varieties.Thirty-seven cultivars could be identified by 4 primer combinations st least.The genetic deversity analyzed indicated that,37 peony varieties could be divided into five types based on the genetic similarity coefficient of 0.69 threshold.
