Abstract:
Taking native species of pod pepper from Lipin county of Guizhou province as materials,cotyledons of germinated aseptic seedlings were cut as explants.Adopting the plant tissue culture method with solid medium,effects of different media of a basal nutrient medium supplemented with different auxins,cytokinins and auxin-cytokinin combinations,different physiological ages of aseptic seedlings and different parts of cotyledon on cotyledon in vitro culture system and plant regeneration,such as in vitro cotyledon inoculation,callus induction,adventitious bud differentiation,bud elongation,root induction,domestication,transplantation and growing adult plant were investigated.The results showed that the optimal medium for adventitious bud differentiation was MS+6-BA 2 mg/L+NAA 0.1 mg/L,with 100% bud differentiation percentage.The optimal medium for bud elongation was MS+KT 2 mg/L,with 50% bud elongation percentage.And the rooting medium was MS without other supplements,with 44% taking root percentage.The average of 15-18 adventitious clustered buds were differentiated by one explant.Among these adventitious buds differentiated per explant,14-16 buds were elongated,and 6-7 seedlings were obtained at last.This period was 50-55 days from cotyledon explant inoculation to regeneration seedlings.The survival percentage of regeneration seedlings after transplanting was 98%.The efficient regeneration system by in vitro culture was preliminarily established on pod pepper of Guizhou province,which laid important foundation for tissue culture in rapid propagation and genetic transformation of pod pepper in this study.
