Abstract:
Taking LiliumtenuifoliuOriental×Tumpet‘Robina’asmaterials,andadventitiousbudswhichdifferentiatedfrombulbscales,filamentsandstalkofitwereusedtoformembryogeniccallus,multiplicationwasconductedviasolid,liquidandliquidpreculturethensolidculturemethods.Theresultsshowedthatitproducedthemostembrogeniccalluswhenexplantswereculturedoninductionmediumwhichcontained0.5mg/LNAA.Thebestresultofcallusproliferationwasmade viaaalternativeculturemethodwhichcalluswastransferredonsolidandliquidmediumbyturns.The GUSgenecontainedinvectorpCAMBIA1301wasintroducedintotheembryogenicculturesbyAgrobacterium-mediatedtracientexpressionandthenGUSchemicaltissuesstainingwasconductedtoobserve.Theresultsshowedthatitwasnosignificanttransientexpressionefficiencyvarianceamongcallusofthreecultivationmethods.
