Abstract:
The indigenous endophytic bacteria in the tissue culture seedlings germinated from seeds of Ziziphus jujuba var.fupingdazao were studied using observation with optical microscope and scanning electronic microscope (SEM),and detection of specific 16S rDNA fragment.The results showed that a large quantity of endophytic bacteria in jujube seedlings were detected under optical microscope with a magnification of 1 000 times,and round,short rod or rod bacterial cells with the size of 1.5~1.8 μm,(2.2~2.9) μm×(1.1~1.9) μm and (3.7~4.4) μm×(1.6~1.9) μm,respectively,were observed.Rod bacteria were found by SEM as well.The 16S rDNA fragment of 1.5 kb was amplified from the total DNA of seedlings of jujube with the bacterial primer pair pf27/1525.
