Abstract:

GenomicDNAofasparagusbeanwasextractedbythemethordofDNAsecurePlantkit.FivefactorssuchasconcentrationofMg2+,dNTPs,TaqDNA,primer,andtemplateDNAwhichwereprimaryfactorfortheorthogonaldesignwasselected,whilethebestcyclesofPCRreactionwasalsoscreenedfromthreedifferenttrements.TheresultsshowedthattheoptimizationforISSR-PCRreactionsystemonasparagusbeanwasasfollows：25μLreactionsystemcontaining1.0mmol/LMg2+，0.2μmol/LdNTP，1.5UTaqDNApolymerase，0.3μmol/Lprimerand45ngDNAtemplet,thewholePCRreactionranfor40cycles.
