Abstract:
JuglansregiaL.endocarpwereemployedasmaterialsinthisreport,becauseJuglansregiaL.endocarphadhighdegreeoflignificationandendocarpwererichinpolysaccharidesandpolyphenolcompounds,theperformancesofthreeextractionmethodsoftotalRNA:CTAB1,CTAB2andRNAplantReagentwerecomparedusingintheJuglansregiaL.endocarp.TheresultsshowedthatthetotalRNAisolatedbythreemethodshadthreebands.ThetotalRNAisolatedbythemethodofCTAB1hadtrailedbands,theOD-260/OD-280valueoftotalRNAwas1.540,theyieldwas75.9μg/g,theRNAintegralityandqualitywerefaulty.ThetotalRNAisolatedbythemethodofCTAB2hadintactbands,butitwascaliginous,theOD-260/OD-280valueoftotalRNAwas1.801,theyieldwas86.1μg/g,butthetimeofRNAisolationwasconsumed.TheOD-260/OD280valueofRNAisolatedbythemethodofRNAplantReagentwas2.055,theyieldwas96.4μg/g,threebandswereclean,steadyandbright,thatprovedthemethodcouldremovepolysaccharidesandpolyphenolcompoundscompletely,itcouldextracthighpurityandcompleteRNAatahighproductivity.ThetotalRNAisolatedbythemethodofRNAplantReagentwashighqualityandproductivity,themethodcompletelywasfitforfurtherbiologicalresearch.
