Abstract:
Taking rhizomes axillary buds of Curcuma attenuata as explants with adding 6-BA,NAA,TDZ different concentration and various combinations of MS basic medium, the optimal conditions of tissus culture and rapid propagation technology about shooting induction, multiple shoot subculture, proliferation, rooting, and seddling cultivation were studied. The results showed that the optimal medium for shooting induction was MS+6-BA 2.0 mg/L+NAA 0.5 mg/L with an inducing rate of 86.7% and rapid growth. The proliferation efficiency of adventitious shoots increased 14.1-fold after subculture for three times on MS medium containing 2.0 mg/L 6-BA and 0.1 mg/L TDZ.Medium containing 1.0 mg/L 6-BA and 0.5 mg/L NAA served as the rooting medium.Over 90% of the plantlets survived when they grow more than 6 cm tall.
