Abstract:
The experiment gets across using ten kinds of the hypocotyl and cotyledon of alfalfa as explants were used to study the effect of different mediums,hormones in different concentrations on callus induction rate and differentiation rate.The results indicated that the Medicago sativa L.cv.Guochan hypocotyl callus differentiation rate was the highest;the best medium for callus induction was the MS salt+UM vitamin,2.0 mg/L 2,4-D+025 mg/L KT,2.0 g/L casein acid hydrolysate+6 g/L Agar+30 g/L sucrose;the best differentiation medium was the MS medium+05 mg/L KT+01 mg/LNAA;the best root medium for plant development was 1/2 MS.
