- 以收集的2个野生秀珍菇菌株(YXZ-03和YXZ-07)为试材,采用农艺性状测定、全基因组重测序技术的方法对2个菌株进行鉴定,研究了2个菌株的农艺性状及变异位点,以期为秀珍菇菌株的遗传差异、系统发育分析等提供借鉴,丰富研究手段,同样为后续秀珍菇分子辅助育种、功能基因发掘与机理探索等研究提供参考依据。结果表明:2个菌株性状之间存在差异;全基因组重测序结果进一步表明,2个菌株鉴定出大量的SNP和InDel变异位点。YXZ-03菌株检测到1 755 412个SNP变异位点、259 731个InDel变异位点;YXZ-07菌株检测到1 767 908个SNP变异位点、262 446个InDel变异位点。基于InDel变异序列设计引物,筛选获得2个可以鉴别YXZ-03和YXZ-07菌株的分子标记。
- Abstract:
- Taking two wild Pleurotus pulmonarius strains (YXZ-03 and YXZ-07) as the test materials,and the two strains were identified by agronomics and whole genome resequencing technology.The agronomics and variation loci of the two strains were studied,in order to provide reference for genetic differences and phylogenetic analysis of Pleurotus pulmonarius strains,enrich research methods,and also provide reference for subsequent molecular-assisted breeding,functional gene discovery and mechanism exploration of Pleurotus pulmonarius.The results showed that there were differences between the two strains.The results of whole genome resequencing further showed that a large number of SNPS and InDel variants were identified in the two strains.1 755 412 SNP variants and 259 731 InDel variants were detected in YXZ-03 strain.1 767 908 SNP variants and 262 446 InDel variants were detected in YXZ-07 strain.Primers were designed based on the InDel variant sequence,and two molecular markers were obtained to identify YXZ-03 and YXZ-07 strains.
