- 以在辽宁省锦州市义县大球盖菇种植基地发现红褐色大球盖菇的黄色变异子实体为试材,采用组织分离获得纯菌丝体后,进行初筛、复筛、中间试验和示范栽培的选育方法,以期获得新的大球盖菇种质资源,为大球盖菇的开发利用提供参考依据。结果表明:该试验选育出1株性状优良的黄色大球盖菇菌株“辽金球盖菇1号”,拮抗试验和ISSR分子标记验证表明“辽金球盖菇1号”与出发菌株具有明显的遗传差异,是区别于出发菌株的新菌株。该菌株菌盖金黄色,子实体单菇质量25~90 g;菌丝生长最适温度为25 ℃,最佳碳源为可溶性淀粉,最佳氮源为大豆蛋白胨,最佳无机盐为磷酸二氢钾。
- Abstract:
- The yellow mutant fruiting body of the reddish brown Stropharia rugosoannulata discovered at the Yixian cultivation base in Jinzhou City,Liaoning Province was used as the test material.The breeding method of obtaining pure mycelium through tissue isolation,initial screening,re-screening,intermediate experiments,and demonstration cultivation were carried out,in order to obtain new strain of Stropharia rugosoannulata and provide reference for the development and utilization of Stropharia rugosoannulata.The results showed that a yellow strain of Stropharia rugosoannulata with excellent traits,‘LiaoJQ01’,was selected in this experiment.The antagonistic experiments and ISSR analysis indicated that ‘LiaoJQ01’ and its original strain were different in genetics,and the strain ‘LiaoJQ01’ was a new strain.The cap of the fruiting body of strain ‘LiaoJQ01’ was bright yellow,and the weight of each fruiting body was of 25-90 g.The optimal temperature for mycelial growth of this strain was 25 ℃,the optimal carbon source was soluble starch,the optimal nitrogen source wad soy peptone,and the optimal inorganic salt was potassium dihydrogen phosphate.
